Lilly Endowment is a private philanthropic foundation created in 1937 by J. K. Lilly Sr. and his two sons, Eli Lilly and J. K. Lilly Jr., using gifts of stock in the family's pharmaceutical company, Eli Lilly and Company, rather than a separate cash fortune built apart from the business that made the family's name. The three men established the foundation in Indianapolis, where the pharmaceutical company itself was headquartered, giving the Endowment a lasting regional identity in Indiana alongside its national philanthropic reach.

Following the founders' own stated wishes, religion is one of the Endowment's three core giving areas, alongside education and community development, and its religion grantmaking is aimed principally at strengthening American Christian congregational life and the pastoral and lay leadership that sustains it over time, a focus the foundation has maintained consistently since the Lilly family first set its giving priorities in 1937. The gift of company stock rather than cash tied the Endowment's own long term fortunes directly to the pharmaceutical company's own performance in the decades that followed, an arrangement that has shaped the scale of its later grantmaking far more than any single annual fundraising effort could have.
