Lilly Endowment is a private philanthropic foundation created in 1937 by J. K. Lilly Sr. and his two sons, Eli Lilly and J. K. Lilly Jr., using gifts of stock in the family's pharmaceutical company, Eli Lilly and Company, rather than a separate cash fortune built apart from the business that made the family's name. The three men established the foundation in Indianapolis, where the pharmaceutical company itself was headquartered, giving the Endowment a lasting regional identity in Indiana alongside its national philanthropic reach.

Following the founders' own stated wishes, religion is one of the Endowment's three core giving areas, alongside education and community development, and its religion grantmaking is aimed principally at strengthening American Christian congregational life and the pastoral and lay leadership that sustains it over time, a focus the foundation has maintained consistently since the Lilly family first set its giving priorities in 1937. The gift of company stock rather than cash tied the Endowment's own long term fortunes directly to the pharmaceutical company's own performance in the decades that followed, an arrangement that has shaped the scale of its later grantmaking far more than any single annual fundraising effort could have.

Lilly Endowment's own most recently filed Form 990-PF, the annual regulatory filing the Internal Revenue Service requires of every private foundation, read here through ProPublica's Nonprofit Explorer digitisation of that filing, reports total revenue of about 5.13 billion dollars against total expenses of about 3.96 billion dollars for the fiscal year ended 31 December 2025, with total assets of roughly 105.9 billion dollars, a scale that places it among the very largest private foundations operating in the United States today.

The bulk of that enormous asset value reflects the market price of the Endowment's own concentrated holding of Eli Lilly and Company stock, inherited directly from its 1937 founding gift, rather than a diversified investment portfolio built up independently of the family company over the decades since. As a private foundation under United States tax law, Lilly Endowment is required to file that Form 990-PF publicly every year, making its finances, including its religion grantmaking, a matter of public record rather than private disclosure known only to its own trustees.
